Abstract:Althoughthereareadvancesindiagnostic,predictive,andtherapeuticstrategies,discoveringproteinbiomarkerforearlydetectionisrequiredforimprovingthesurvivalrateofthepatientswithendometrialcarcinoma.Inthisstudy,weidentifyproteinsthataredifferentiallyexpressedbetweentheStageIendometrialcarcinomaandthenormalpericarcinoustissuesbyusingisobarictagsforrelativeandabsolutequantitation(iTRAQ)-basedproteomicanalysis.Totally,wescreened1,proteins.Amongthem,proteinsweresignificantlyoverexpressed,and30weresignificantlydownexpressedinendometrialcarcinoma.Usingthebioinformaticsanalysis,weidentifiedalistofproteinsthatmightbecloselyassociatedwithendometrialcarcinoma,includingCCT7,HSPA8,PCBP2,LONP1,PFN1,andEEF2.WevalidatedthegeneoverexpressionofthesemoleculesintheendometrialcarcinomatissuesandfoundthatHSPA8wasmostsignificantlyupregulated.WefurthervalidatedtheoverexpressionofHSPA8byusingimmunoblotanalysis.Then,HSPA8siRNAwastransferredintotheendometrialcancercellsRL-95-2andHEC-1B.ThedepletionofHSPA8siRNAssignificantlyreducedcellproliferation,promotedcellapoptosis,andsuppressedcellgrowthinbothcelllines.Takentogether,HSPA8playsavitalroleinthedevelopmentofendometrialcarcinoma.HSPA8isacandidatebiomarkerforearlydiagnosisandtherapyofStageIendometrialcarcinoma.
